Description
17-DMAGisaninhibitorofHSP90thatexhibitsneuroprotective,anticancerchemotherapeutic,andantiviralactivities.InanimalmodelsofMachado-Josephdisease,17-DMAGincreaseslevelsofbeclin-1andLC3-11,inducingautophagyandimprovingmotordysfunction.Inneuroblastomacells,thiscompoundexertseffectsonanaplasticlymphomakinase(ALK)andN-Myc,inducingcellcyclearrestandapoptosis.Additionally,17-DMAGinhibitscellgrowthandinducesapoptosisinadultT-cellleukemia(ATL)cells.InanimalmodelswithATLxenografts,17-DMAGinhibitsmetastasisandimprovessurvivalrates.17-DMAGalsopreventsviralproductionbyhumanT-lymphocyticvirustype-1.Inseparatemodels,17-DMAGdecreasesexpressionofconservedherpesvirusproteinkinasesandsuppressesviralgrowth.
