7,8-Dihydroxyflavone(DHF)isabrain-derivedneurotrophicfactor(BDNF)mimeticthatactivatesTrkBreceptors.DHFdisplaysneuromodulatory,neuroprotective,antipsychotic,anti-obesity,anticancer,antioxidative,anti-inflammatory,andantihypertensiveactivities.Inanimalmodelsoftraumaticbraininjury(TBI),DHFdecreasesbrainedema,neuronaldeath,andbraintissuedamageandimprovesfunctionaldeficits.Inanimalmodelsofschizophrenia,thiscompoundreversescognitivedeficitsandpromotessynapticplasticity.DHFalsobenefitsneurodegenerativediseasemodelssuchasParkinson’sdisease,Alzheimer’sdisease,andamyotrophiclateralsclerosis(ALS);inthesemodels,DHFdecreasesmotordeficitsandimprovescognitiveABIlities.DHFpreventstheinductionofdiet-inducedobesityinanimalmodelsandalsodecreasesADIposity,increasesenergyexpenditure,andimprovesinsulinsensitivityinalready-obeseanimals.Inoralsquamouscellcarcinomacells,thiscompounddecreasescellgrowthandinducesapoptosisbysuppressingSp1signaling.DHFalsodecreasesproductionofNOandPGE2andsuppressesexpressionofCOX-2,iNOS,TNF-α,andIL-1βinLPS-stimulatedmicroglia.Inothercellularmodels,DHFincreaseslevelsofHO-1andNrf2,protectingagainstH2O2-andUVlight-inducedoxidativedamage.DHFalsoincreaseseNOSexpressioninvitroanddecreasesbloodpressureinspontaneouslyhypertensiverats.
