Description

CandesartanisaninhibitoroftheangiotensinIItype1receptor(AT1)thatdisplaysantihypertensive,cardioprotective,nephroprotective,andpotentiallyantiviralactivities.Invivo,candesartanincreasesrenalbloodblowanddecreasesrenalvascularresistance,glomerularfiltrationrate,andfiltrationfraction.CandesartanalsodecreasesNADPHoxidaseactivityandlevelsofTGF-β,inhibitingcalciumoxalatecrystaldepositionandkidneystoneformation.Inanimalmodelsofpressureoverload-inducedcardiacremodeling,candesartandownregulatesSmad3andfibronectin,upregulatesSmad7,andinhibitsmatrixmetalloproteinase9(MMP9)andtheepithelial-to-mesenchymaltransition(EMT),preventingcollagendeposition,leftventricularremodeling,anddecreasesinleftventricularejectionfraction.Additionally,thiscompounddownregulatesexpressionofVEGFR2,decreasingretinalneovascularizationwithoutinhibitingtotalangiogenesis.Separately,candesartanmayinhibittheinteractionbetweenlensepithelium-derivedgrowthfactor(LEDGF)andHIV-1integrase,exhibitingpotentialasatreatmentforHIV.
