Description
Bortezomibisaproteasomeinhibitorthatdisplaysanticancerchemotherapeuticandantiviralactivityinvitro,invivo,andinclinicalsettings;itisclinicallyusedorintrialsfortreatmentofglioma,acutelymphoblasticleukemia(ALL),mantlecelllymphoma,andmultiplemyeloma.Ingliomacells,bortezomibinducesactivationofcaspase3andapoptosis;itsinhibitionoftheproteasomestimulatesangiogenesisthroughanincreaseinproductionofVEGFandHIF-1α.Incolorectalcancercells,bortezomibincreasedlevelsofROS,inducingG2/Mphasecellcyclearrest.Additionally,bortezomibinducesneuropathy,potentiallythroughincreasingpolymerizedtubulinlevelsinneurons.InmodelsofRiftValleyFeverVirus,bortezomibdecreasesviralloadbysuppressingnonstructuralS-segmentproteinformationofnuclearfilaments.
