Description

PJ34isaphenanthridinonederivativethatactsasacompetitivepoly(ADP)-ribosepolymerase(PARP)inhibitor.PJ34exhibitscardioprotective,neuroprotective,anticancer,andanti-diabeticactivities.Ininvitroandinvivomodelsofoxygenglucosedeprivation,PJ34protectsagainststroke-relatedischemicbraininjury.Thiscompoundisalsocardioprotectiveagainstmyocardialinfarction,decreasinginfarctsizeandpostsystolicshortening.IndiabeticNODmice,PJ34decreasedinsulitis,beta-celldestruction,anddiabetesincidencebydecreasingisletIFN-γexpressionandinducingapoptosisinislet-infiltratingleukocytes.Inmousemodelsofallergicencephalitis,PJ34exhibitsprotectivebenefitbyshiftingtheimmuneresponsefromapro-inflammatoryTh1-mediatedresponsetoananti-inflammatoryTh4-mediatedresponse.Additionally,inavarietyofcancercelllines,PJ34inducesG2/Mphasemitoticarrestandcaspase-depedentapoptosis,potentiallyduetodirectantagonismofPim-1kinase.
