Description
Thebosutinibstructuralisomerdiffersfrombosutinibitselfonlyinthepositionsofthechlorineandmethoxysubstituentsontheanilinering.BosutinibisananticancerchemotherapeuticcompoundthatinhibitssrcandAblkinasesandisclinicallyusedtotreatchronicmyeloidleukemia.Invivo,bosutinibdoesnotinduceapoptosisbutdoesdecreasecellproliferation,inhibitcelldifferentiation,andinhibittumormetastases,alllikelythroughinhibitionofdownstreamMAPKsignaling.Additionally,bosutinibmayalterIGF-1andIGFbindingprotein3(IGFbp3)signalingaswell.ThiscompoundalsoshowsneuroprotectiveandcognitionenhancingbenefitsinanimalmodelsofAlzheimer’sdisease.Bosutinibclearsβ-amyloidplaquesandincreasescognitivefunctionbyfacilitationinteractionbetweenparkinandBeclin-1proteins,whichthenaccumulateinthelysosomefordegradation.
