Description
Asparaginaseisanenzymethatcatalyzesthehydrolysisofasparaginetoaspartateandammonia,depletingasparaginelevelsandinhibitingcellgrowth;itiscommonlyusedasananticancerchemotherapeuticandanti-angiogeniccompoundinthetreatmentofleukemia.Asparaginaseinhibitsproliferationofacutemyelogenousleukemia(AML)cells,inducingapoptosismediatedbyapoptosis-inducingfactor(AIF).Asparaginasedecreasesβ1integrinbindingandphosphorylationofFAK,inducingautophagy;thisdecreasesmicrovascularendothelialcelltubeformationandinhibitsinvasionofovariancancercells.Inclinicalsettings,asparaginasemayinducethromboticeffect,asadmiNISTrationoftenresultsindepletionofanti-thrombin,anditmayalsoincreaseserumtriglycerides.
