Description

FluvastatinisaninhibitorofHMG-CoAreductasethatisclinicallyusedtolowercholesterolandtreatcardiovasculardisease.Fluvastatinexhibitsanti-hyperlipidemic,anti-fibrotic,cardioprotective,antiviral,antithrombotic,antioxidative,vasorelaxant,anti-atherosclerotic,andanticanceractivities.FluvastatinsuppressesviralloadofhepatitisCvirusinaclinicalsettingandpreventsviralreplication.Invitro,fluvastatindecreasesplateletactivationbyincreasingPECAM-1signalingandinhibitingactivationofAkt.Additionally,fluvastatinscavengeshydroxylrADIcalsinvitro.Inothercellularmodels,fluvastatinincreasesexpressionofNOandphospholipase2(PLA2)anddecreaseslevelsofangiotensinII(ATII)andROCK.Additionally,thiscompoundinducesapoptosisandG2/Mphasecellcyclearrest,decreasesthemitochondrialmembranepotential,andincreasesreleaseofcytochromecandactivationofcaspase3inhepatocellularcarcinomacells.Inanimalmodelsofcardiacdistress,fluvastatinimprovesleftventricularfunctionandpreventfibrosisbyinhibitingRhoAanddecreasinglevelsofCTFGandfibronectin.
