Description
Dehydrocostuslactone(DHL)isasesquiterpenelactoneoriginallyfoundinspeciesofSaussurea.DHLexhibitsantibiotic,antioxidative,anti-angiogenic,anticancerchemotherapeutic,andchemopreventiveactivities.DHLdisplaysantibacterialactivityagainstspeciesofMycobacterium.Inosteoblasts,DHLincreaseslevelsofNADPH,thioredoxinreductase,andphospho-CREBanddecreaseslevelsofnitrotyrosin,preventingmitochondrialdysfunctioninducedbyantimycinA.InmacrophagestreatedwithLPS,DHLdecreasesexpressionofiNOSandTNF-α.Additionally,DHLactivatescAMP-activatedcysticfibrosistransmembraneconductanceregulator(CFTR)Cl-channelsinthyroidcells.Invitroandinvivo,thiscompounddownregulatesexpressionofcyclinD1,Akt,mTOR,andinhibitsphosphorylationofGSK-3β,inducingG0/G1phasecellcyclearrestandpreventingcapillarytubeformation.Insofttissuesarcomacelllines,DHLdecreasesexpressionofCDK2,p27,CDK1/cdc2,cyclinB1,andmatrixmetalloproteinases2and9(MMP2/9),inducingG2/Mphasecellcyclearrestandinhibitingcellproliferation.
