Description

NSC-74859isaninhibitorofpSTAT3thatexhibitsanticancerchemotherapeutic,antiviral,anti-fibrotic,andnephroprotectiveactivities.NSC-74859enhancesthetoxicityofdoxorubicinbyinhibitingtheepithelial-to-mesenchymaltransition(EMT)invitro;italsoenhancestheefficacyofcetuximabinhepaticcarcinomacells.Incellularandanimalmodelsofhepatocellularcancer,NSC-74859suppressescellproliferationandinhibitstumorgrowthofxenografts.InhibitionofpSTAT3resultsindecreasedexpressionofgenesencodingcyclinD1,Bcl-xl,andsurvivin.Additionally,NSC-74859inhibitsreplicationofvaricella-zostervirus(VZV),aherpesvirus.Inanimalmodelsofkidneyinjury,NSC-74859decreasesinfiltrationofinflammatorycells,decreasesexpressionoffibronectin,smoothmuscleactin,andcollagentype-1proteins,andinhibitsproliferation;together,thisincreasesapoptosisinrenalinterstitialfibroblasts.
