ParthenolideisasesquiterpenelactonefoundinTanacetumthatexhibitsanticancerchemotherapeutic,anti-metastatic,anti-angiogenic,anti-inflammatory,andantinociceptiveactivities.ParthenolideactsasapartialagoNISTattransientreceptorpotentialankyrin1(TRPA1)channelsanddesensitizesthem,preventingreleaseofcalcitoningene-relatedpeptide(CGRP).Additionally,parthenolideinhibitsATPaseactivityofNLRP3andproteaseactivityofcaspase1.Inmultiplemyelomacells,parthenolidedecreasesexpressionofNF-κB,VEGF,andIL-6andincreasesexpressionofIκBkinase,inhibitingcellmigrationandtubuleformation.Innon-smallcelllungcancer(NSCLC)cells,parthenolidedecreaseslevelsofMCL-1andincreaseslevelsofMAIP-1,triggeringERstressandinducingcellcyclearrestandapoptosis.Inbreastcancercells,thiscompoundactivatesNADPHoxidaseandincreasesROSgeneration,increasinglevelsofp-JNKanddownregulatingNF-κB,VEGF,andmatrixmetalloproteinases2and9(MMP2/9);invivo,parthenolideinhibitstumorgrowthandmetastasis.
