Description
Pixantroneisananticancerchemotherapeuticandimmunosuppressiveaza-anthracenedionethatactsasaDNAintercalatorandtopoisomeraseIIinhibitor;thiscompoundintercalatesviathemajororminorgroovesofDNA,bindingtoCpGorCpAmotifs.Incellularmodels,pixantroneinhibitsproliferationofTcells,butdoesnotaffectdifferentiationofdendriticcellsorBcells.Inclinicaltrials,pixantroneincreasessurvivaltimesofpatientswithrelapsedorrefractoryaggressivenon-Hodgkin’sB-celllymphoma.Pixantronealsoshowspotentialbenefitinthetreatmentofseveralotherdiseases.Incellularmodels,thiscompounddecreasesorpreventsamyloid-β(Aβ)oligomerization,indicatingapotentialuseinthetreatmentofAlzheimer’sdisease.Additionally,itdecreasesacetylcholinereceptor(AChR)-specificimmuneresponsesandpreventsandtreatsexperimentalautoimmuneencephalitis(EAE)inanimalmodels,suggestingpotentialbenefitinthetreatmentofmyastheniagravisandmultiplesclerosis.
