PropofolisanagoNISTatGABA-Areceptorsandantagonistatvoltage-gatedNa+channelsandNMDAreceptors;italsomodulatesCa2+signaling.Propofoldisplaysanesthetic,sedative/hypnotic,neuroprotective,andnephroprotectiveactivities.Propofolisashort-actingintravenousanestheticthatdepressesthecentralnervoussystem,decreasescerebralbloodflow,anddecreasesintracranialpressure.Inamiddlecerebralarteryocclusion(MCAO)modelofcerebralischemia/reperfusion,propofoldecreasesexpressionofaquaporins1and4(AQ-1andAQ-4),matrixmetalloproteinases2and9(MMP2/9),andHIF-1α,decreasingedemaanddisruptionoftheblood-brainbarrier.Propofolalsoincreasesexpressionofμ-opioidreceptorsinneuroblastomacells.Additionally,thiscompoundnormalizesosmolality,serumcreatine,andlevelsofAQP-2,ICAM-1,TNF-α,Bcl-2,andBaxinkidneys,improvingrenalfunctioninanimalmodelsofendotoxemia-inducedkidneyinjury.
