Description
Quetiapineisasecond-generationatypicalantipsychoticthatisusedclinicallytotreatbipolardisorder,schizophrenia,anddepression.QuetiapineinhibitsD1/2/3/4receptors,5-HT2A/2C/6/7receptors,histamineH1/2receptors,M1muscarinicacetylcholinereceptors(mAChRs),andα1A/1B/2C-adrenergicreceptorsandactivatesσ1/2receptorsand5-HT1Areceptors.Quetiapine’sneuromodulatory,cognitionenhancing,antipsychotic,antidepressant,antihistamine,andneuroprotectiveactivitiesstemfromitsactionsatD2,5-HTandhistamineH1/2receptors.Themajormetaboliteofquetiapine,norquetiapine,alsoexhibitssimilaractivitiesbuthasadifferentbindingprofile.WhenadmiNISTeredinvivo,quetiapinedecreasesimmobilitytimeintheforcedswimtest.Quetiapinealsoincreasesoligodendrocytematurationandpreventslossofoligodendrocytesandmyelininanimalmodelsofcerebralischemia/reperfusion.QuetiapinealsoexhibitsbenefitinthetreatmentofAlzheimer’sdisease,suppressingmemoryimpairmentintheY-mazetestintransgenicanimalmodelsanddecreasingage-relatedlossofbrain-derivedneurotrophicfactor(BDNF).
