N-Acetyl-S-(N’-phenylhexylthiocarbamoyl)-L-cysteine(PHITC)isacysteineconjugateofN-acetyl-phenylhexylisothiocyanate.IsothiocyanatesaretypicallyfoundinplantsoftheBrassicaceaefamily,includingbroccoli,cabbage,andrADIsh.Isothiocyanatesarebestknownfortheirantioxidative,anticancerchemotherapeutic,chemopreventive,anti-angiogenic,andantibioticproperties.Invitro,PHITCincreasescaspase3activityandcleavageofpoly(ADP)-ribosepolymerase(PARP),inducingcaspase-mediatedapoptosisincellularmodels.ThiscompounddecreasesoxidationofcarcinogenNNKandincreasesactivityofNADPH:quinoneoxidoreductaseandglutathione-S-transferaseinvitroandinvivo.PHITCinhibitshistonedeacetylase(HDAC)activity,IL-6receptorproduction,andVEGFexpression;italsoinducesmitochondrialmembranedepolarizationandapoptosisinmyelomacells.Inleukemiacells,PHITCinhibitsexpressionofcyclinandincreasesexpressionofCDKinhibitors,inducingapoptosisandinhibitinggrowthofxenograftsofthesecellsinanimals.
