Description
Lenalidomidedisplaysanti-inflammatory,anti-angiogenic,immunosuppressive,andanticancerchemotherapeuticbenefitsinthetreatmentofmultiplemyelomaandmyelodysplasticsyndromesassociatedwithchromosome5qdeletions.Lenalidomideisaderivativeofthalidomideandisalsocurrentlyinclinicaltrialsasapotentialtreatmentforseverallymphomasandleukemias;likethalidomide,itmayinhibitTNF-αandcereblon.Invitro,lenalidomideupregulatesbothBH3-interactingdomaindeathagoNIST(BID)andFOSanddownregulatedNKX2-1,increasingapoptosisandinhibitingcellproliferation.Additionally,thiscompoundinhibitsVEGF-inducedPI3K-AktsignalingaswellasexpressionofHIF-1α.
